Tadalafil is a phosphodiesterase type-5 (PDE5) inhibitor in the same family as sildenafil (Viagra) and vardenafil (Levitra). Its standout characteristic is a very long elimination half-life of approximately 17.5 hours, compared to about 4 hours for sildenafil — a single dose can remain clinically active for over 36 hours, which is where the "weekend pill" nickname comes from.



Mechanistically, Tadalafil inhibits the breakdown of cyclic GMP in vascular smooth muscle, supporting blood flow in response to natural stimulation.
